ABSTRACT

【Objective】 To establish a platelet donor database with HPA and HLA high resolution results in Hefei area, and to analyze the appropriate pool capacity, so as to provide support for patients with immune PTR transfusion difficulties. 【Methods】 The HPA-1-6, -10, -15 and 21 genotypes of 1157 voluntary blood donors in Hefei were analyzed by multiplex PCR and the frequencies were calculated. HLA-A, -B high resolution gene frequency and haplotype frequency in 1115 voluntary blood donors in Hefei were calculated using maximum expected value method. 【Results】 Hefei database of platelet donors was established according to the analysis of HPA and HLA high-resolution gene polymorphisms in Hefei population.In the donor pool with 1 157 cases of known genotypes HPA-1-6, -10, -15, -21, about 91% of patients can find an donor with full-matched HPA genotype. In the donor pool with 1 115 cases of known HLA-A, -B high-resolution genotype, the probability of finding at least one identical donor is about 60%. 【Conclusion】 The local-suitable database of platelet donors with known HPA and HLA genotypes in Hefei has been established. After analysis, this database basically meets the needs of patients with immune PTR to find suitable platelets.

ABSTRACT

Objective: To observe the effect of phenylephrine (PE) on pressure overload induced myocardial ifbrosis (MF) with its relevance to transforming growth factor-β1 (TGF-β1), drosophila mothers against decapentaplegic protein 3 (smad3) and connective tissue growth factor (CTGF) signal pathway in experimental rats. Methods: A total of 28 male SD rats were randomly divided into 4 groups: Control group, AAC (abdominal aorta coarctation) group, AAC+PE group and AAC+prazosin group.n=7 in each group. Collagen volume fraction (CVF) of left ventricle was observed by myocardial collagen morphology, left ventricular myocardial tissue protein expressions of α-smooth muscle actin (α-SMA), TGF-β1, smad3 and CTGF were measured by immunohistochemistry, protein expression of α-SMA was also examined by Western blot analysis. Results:①Myocardial collagen morphology presented that compared with Control group, AAC, AAC+PE and AAC+prazosin groups had increased CVF, allP Conclusion: Phenylephrine could improve pressure overload induced MF in experimental rats which might be related to TGF-β1/smads signal pathway inhibition and CTGF down-regulation.

ABSTRACT

Objective:To explicit phenylephrine (PE), α1-adrenergic receptor (α1-AR) on myocardial fibrosis regulation and interleukin-1 (IL-1), IL-6, tumor necrosis factor-α (TNF-α) expressions in pressure overloaded mice. Methods:A total of 49 KM mice were randomly divided into 3 groups: Blank control group, n=7, Sham operation group, n=7, Transverse abdominal aortic constriction (TAC) group, n=35, and 8 weeks later, the mice in TAC group were further divided into 5 sub-groups as TAC control, TAC+PE, TAC+Praz, TAC+Prop, TAC+Carv sub-groups, n=5 in each sub-group, and the animals were respectively treated for 3 weeks. Left ventricular collagen volume fraction (CVE), hydroxyproline content and IL-1, IL-6, TNF-α expressions were examined respectively. Results: By 8 weeks treatment, compared with Blank control group, TAC group had obvious myocardial fibrosis, increased hydroxyproline content and IL-1, IL-6, TNF-α expressions,P0.05; CVE and hydroxyproline content were similar between TAC+PE and TAC+Prop sub-groups, while the expressions of IL-1, IL-6 and TNF-α were obviously decreased in TAC+Prop sub-group,P<0.05. Conclusion: PE may improve myocardial ifbrosis and IL-1, IL-6, TNF-α expressions by activating α1-AR in pressure overloaded mice, α1-AR might be a defending factor for myocardial ifbrosis.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of Dendrobium officinale granule (DOG) on symptoms, blood pressure and serum biochemical indexes of long-term-alcohol-induced hypertension rats.</p><p><b>METHOD</b>The alcohol-induced hypertension rat model was established by feeding alcohol drink to normal rats (the alcohol volume fraction increases from 5% to 22%). Since the 4th week, DOG was administered for 32 weeks, once everyday. During the experiment, body weight, kinematic parameters (locomotor activities, grip strength, duration of vertigo) and blood pressures (systolic blood pressure, diastolic blood pressure and mean blood pressure) were detected regularly. On the 28th and 32nd weeks, blood samples were collected to determine serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), uric acid (UA), creatinine (Cr), cholesterol (CH) and triglycerides (TG).</p><p><b>RESULT</b>(1) Sign: The DOG-administered group showed reduction in the duration of vertigo and increase in appetite, body weight, locomotor activities and grip strength. (2) Blood pressure: The DOG-administered group showed significant decrease in blood pressure since the 8th week. (3) Biochemical indexes: The DOG-administered group showed notable decrease in serum ALT, AST, ALP, Cr, UA, TG level, but without significant change in TC level.</p><p><b>CONCLUSION</b>The long-term administration of DOG can relieve alcohol-induced hypertension, while alleviating general signs, liver and kidney injuries and abnormal blood fat biochemical indexes.</p>

ABSTRACT

Objective Calcineurin-NFAT pathway in the regulation of VSMCs proliferation induced by catecholamines.Methods Primary VSMCs from rat aorta were used as the experimental model.Proliferation of VSMCs was measured by MTT assay and cell count.Calcineurin protein and its activity were assayed with immunoblotting and free inorganic phosphate content analysis respectively.Localization of NFATc1 was detected by immunofluorescence staining.Results Phenylephrine(PE,an ?1-adrenoceptor agonist) increased VSMCs proliferation.Prazosin(an ?1-adrenoceptor antagonist),cyclosporin A(CsA,an inhibitor of calcineurin) and chelerythrine(an inhibitor of PKC) decreased PE-induced absorbance and cell number.Timolol(?-adrenoceptor antagonist) has no effect on absorbance and cell number induced by PE.Additional treatment with CsA further inhibited PE-induced absorbance and cell number compared with the chelerythrine pretreatment group.CsA and chelerythrine alone had no significant effect on either absorbance or cell number.CsA decresed PE-induced alcineurin level and its activity.NFATc1 was translocated from cytoplasm to nucleus upon treatment with PE.This translocation was reversed by CsA.Conclusion CsA partially inhibits PE-induced VSMCs proliferation via inhibiting calcineurin activity and NFATc1 nuclear translocation.Calcineurin-NFATc1 pathway is involved in hyperplastic growth of VSMCs induced by catecholamines.
